This study aims to elucidate the mechanism of cardiotoxicity caused by the off-target effects of the antiviral drug remdesivir. Previous research has shown that remdesivir activates vasoconstriction-related Urotensin II Receptor (UTS2R), leading to cardiac dysfunction such as bradycardia and hypotension. Despite these physiological insights, the underlying structural basis of this interaction has not yet been explored._x000D_

By integrating cryo-electron microscopy (cryo-EM) for structural analysis, molecular dynamics (MD) simulations, and cell-based assays, this research seeks a comprehensive understanding of the activation mechanism. These findings will contribute significantly to designing drugs that avoid side effects and establishing a foundation for treating cardiovascular diseases.

Message

GPCRs are major drug targets, but some cause unexpected side effects. I want to elucidate this unexpected activation via this study to develop a methodology to avoid side effects.
